Plus ones specify that they can be used during *Action Rolls* to add 1 to the result. In this case the Spell specifies that you roll your base dice (which means the dice on your board, excluding item bonuses), and the higher roller wins. If you chose a Hero as one Character, can they use a +1 to add to their roll? In any case the caster becomes the defender there for the remainder of the round. The spell triggers after the attack has found its targets but right before damage is actually taken, so the newly added casting hero would not become victim to further AOE damage by standing in the tile being hit, they will only take the damage for the character they are casting on. The caster could use this spell to fly in and take the damage instead of one of those heroes, though they could not prevent the whole attack, and any other heroes would still take damage as normal. In the case of an AOE attack, the attack could harm a number of heroes in a tile. The caster cannot use further means of preventing this damage.
